RESUMO

Objective:To investigate the protective effects of overexpression of long-chain noncoding RNA FAM224B on lung tissue of rats with severe pneumonia and the underlying mechanism.Methods:From August 2020 to March 2021, we randomly allocated 20 rats into the pneumonia group (severe pneumonia modeling) and FAM224B group (severe pneumonia modeling + FAM224B plasmid), with 10 rats in each group. We performed a quantitative real-time polymerase chain reaction to detect the level of FAM224B in lung tissue and performed an enzyme-linked immunosorbent assay to detect the levels of tumor necrosis factor-alpha, interleukin-6, and interleukin-1β in lung tissue. We used the software starBase v2.0 to predict the target gene of FAM224B. We performed a quantitative real-time polymerase chain reaction to detect the expression of the target gene in lung tissue and performed a western blot assay to detect the protein expression of the nuclear factor-kappa B signal pathway in lung tissue.Results:FAM224B expression was (1.09 ± 0.23) and (10.12 ± 1.52) in the pneumonia and FAM224B groups, respectively. FAM224B expression was significantly lower in the pneumonia group compared with the FAM224B group ( t = 15.86, P < 0.01). The levels of tumor necrosis factor-alpha, interleukin-6, and interleukin-1β were (41.53 ± 2.46) μg/L, (34.01 ± 2.53) ng/L, (20.92 ± 1.95) μg/L in the pneumonia group and they were (21.71 ± 2.25) μg/L, (17.13 ± 3.01) ng/L, (11.97 ± 1.21) μg/L in the FAM224B group. There were significant differences in the levels of tumor necrosis factor-alpha, interleukin-6, and interleukin-1β between the two groups ( t = 15.94, 14.29, 13.89, all P < 0.01). FAM224B had complementary binding sites with miR-34b-5p. The expression level of miR-34b-5p in lung tissue was significantly lower in the FAM224B group compared with the pneumonia group ( t = 15.55, P < 0.01). The protein expression levels of phosphorylated nuclear factor-κB subunit (p-p65) and phosphorylated inhibitor of kappa B alpha in lung tissue were significantly lower in the FAM224B group compared with the pneumonia group. Conclusion:FAM224B overexpression reduces the inflammatory reaction in lung tissue of rats with severe pneumonia through inhibiting miR-34b-5p expression.

Objective To explore the effect of statin in treatment of hypertensive atherosclerotic patients with cerebral infarction and the its effect in prevention cerebral infarction recurrence.Methods 108 hypertension patients with atherosclerotic cerebral infarction were randomly divided into two groups,each group had 54 cases.The control group received conventional treatment;the observation group was given combination therapy with statin and the conventional treatment.The efficacy was evaluated after 2 months.The blood pressure,blood lipid and cerebral function,cerebral infarction recurrence within 1 year after treatment in the two groups were observed.The clinical efficacy of the two groups was compared.Results After treatment,blood pressure,blood lipid indexes and brain function of the observation group were better than those of the control group(all P < 0.05).The total effective rate of the observation group was 100%,which was higher than 76% in the control group(x2 =9.488,P < 0.05).The control group had 12 cases of recurrence(recurrence rate 22%),1 case recurrence in the observation group(recurrence rate 2%),with no significant difference between two groups (x2 =13.277,P < 0.05).Conclusion The clinical efficacy of statins in the treatment and prevention of hypertension atherosclerotic cerebral infarction is safe and significant,which is worth to use in clinical practice.

RESUMO

Objective To optimize the technological conditions of polysaccharide extraction from Elaeagnus angustifolia L. by enzymatic-assisted ultrasonic. Methods The effects of different enzymes, enzymolysis temperature, enzyme dosage, enzymolysis time, material-water ratio, crushing degree, ultrasonic time and ultrasonic temperature on the extraction rate of Elaeagnus angustifolia L. polysaccharide was analyzed. Results The optimal technological condition was as follows:1.5% cellulase, 1.5% pectic enzyme, enzymolysis time 40 min, enzymolysis temperature 55 ℃, ultrasonic time 25 min, ultrasonic power 400 W, material- water ratio 1∶30, and temperature 60 ℃. In these conditions, the extraction efficiency of polysaccharide was 12.35%. Conclusion The process is simple and practicable, and can be used for the extraction of Elaeagnus angustifolia L. polysaccharide.

RESUMO

Objective To study the optimum flavonoids extraction process of Elaeagnusangustifolia L. with ethanol. Methods The best condition of extraction process of flavonoids was investigated by orthogonal design. Results The ratio of extraction on this condition was 2.295 mg/g. Conclusion The optimum technological conditions attained by theorthogonal experiment was as follows:concentration of ethanol 70%, extraction temperature 45 ℃, extraction time 30 min and the ratio of Elaeagnusangustifolia L. to extractant 1∶16.

Objective To investigate the age-related change of secretory leukocyte protease inhibitor (SLPI) content in stimulated whole saliva. Methods Ninety-four healthy adults aged 20-94 yrs were enrolled. Samples of whole stimulated saliva were collected from all subjects, the SLPI content was analyzed by enzyme-linked immunosorbent assay (ELISA). <p><b>OBJECTIVE</b>To investigate the changes of cytokeratin 19 during oral carcinogenesis.</p><p><b>METHODS</b>53 specimens including normal oral mucosa, oral epithelial hyperplasia, oral epithelial dysplasia and oral squamous cell carcinoma were investigated by immunohistochemistry, SDS-PAGE and Western blotting.</p><p><b>RESULTS</b>CK19 was detectable in suprabasal cell layers in epithelial dysplasia and in oral cancer, especially in poor-differentiated cancerous cells. With the lesions getting worse, the positive rate, the intensity and the constituent ratio of CK19 raised significantly.</p><p><b>CONCLUSIONS</b>The results suggest that the CK19 expression in suprabasal cell layers of oral mucosa can be used as a marker of diagnosis of oral precancerous lesions and CK19 expression is the initial events during oral carcinogenesis.</p>

RESUMO

Objective:To determine the effectiveness of the interve ntion of burning mouth syndrome(BMS) reported by Chinese articles and to assess the quality of the studies by evidence-based medicine. Methods: relevant literatures were searched and identified in Chinese Medical Bio logical Database (CMB) and Chinese Technological Periodical Database (VIP) . Stu dies were selected with following criteria: study design- randomized controlle d trials; participants-patients with BMS. An evidence-based analysis was perfo rmed to the literatures. Results: There were totally 47 articles examined and only 11 of them met the criteria . Meta analysis was not possible due to the low quantity and poor quality, However, as far as the p ublished articles concerned, Lusunjing capsule showed good short-term effect in the treatment and nerve block with vitamin B 1 and B 12 was more effec tive than oral admenistering of vitamin. Conclusion: Th e quantity and quality of the reviewed studies are far from satisfaction and ca n not meet the clinical needs. Further trials of high methodological quality ne ed to be undertaken in order to develop effective treatment of BMS.
